Utilization of Immunomagnetic Separation for Detection of Salmonella in Raw Broiler Parts
نویسندگان
چکیده
This study was conducted aiming to compare the conventional microbiological method to detect Salmonella in broiler parts with the Immunomagnetic Separation method (IMS) followed by plate isolation and also the IMS associated with Rappaport-Vassiliadis broth (RV). The IMS was performed following a preenrichment step in buffered peptone water. Sixty-one samples (raw broiler parts) were tested and the results showed that the use of the IMS method alone allowed the isolation of Salmonella in 9 of the tested samples, while the association IMS/ RV detected the agent in 30 samples. The conventional microbiological method was able to isolate the agent in 25 opportunities. These results allowed to conclude that the IMS/RV association presented an increased sensitivity and permitted a better isolation of Salmonella. The conclusion was that other means of isolation, in particular those which do not interfere with the growth of bead bounded Salmonella, should be searched.

Introduction: Detection of Cryptosporidium and Giardia parasites in water samples is usually performed by US Environmental Protection Agency 1623 method. Nevertheless, the USEPA1623 method still need improvement, to prevent and control the water borne parasitic disease. Therefore, we undertook the present study. Materials and methods: Totally 48 surface water samples were collected. Four sampl...

The poultry industry is now operating under increased regulatory pressure following the introduction of the pathogen reduction and hazard analysis critical control point (HACCP) rule in 1996. This new operation scheme has greatly increased the need for on-farm food safety risk management of foodborne bacteria, such as Salmonella. Information needed to make informed food safety risk management d...
متن کاملEvaluation of Immunomagnetic Separation for the Detection of Salmonella in Surface Waters by Polymerase Chain Reaction
Salmonella spp. is associated with fecal pollution and capable of surviving for long periods in aquatic environments. Instead of the traditional, time-consuming biochemical detection, polymerase chain reaction (PCR) allows rapid identification of Salmonella directly concentrated from water samples. However, prevalence of Salmonella may be underestimated because of the vulnerability of PCR to va...

To identify the reservoirs of shiga toxin-producing Escherichia coli O157, sensitive detection andisolation methods are necessary. The sensitivity of traditional culture methods can be improved significantlyby the inclusion of an immunoconcentration step, resulting in less false-negative results. In this study, enrichment procedure and immunomagnetic separation (IMS) were compared for use in con...
